Upsample after matching on binned images
Hey folks!
I am using the cisTEM_pvalue_250530 version for 2DTM.
Initially I matched on bin2 images for speed and everything worked accordingly.
Subsequently, the extraction and reconstruction from binned images also worked as expected when run through the cisTEM GUI.
Now, I am interested in extracting from the original, unbinned images.
Based on some of the recent methods sections using 2DTM, I thought that I could replace the binned image path to point to the unbinned images and just specify the smaller pixel size (0.9286 A) during creation of the Refinement package from the MT results.
Extraction did proceed using the Refinement Package creation GUI, but the Contained Particles list still shows the binned pixel size (1.8572 A) and attempts at reconstruction from this extraction result in noise.
Is there something that I need to edit in the database in addition to switching the micrograph image paths to force use of the unbinned images correctly?
Thanks for the help,
-Tom
Good News!
Good news everyone!
Modifying the image file path AND the pixel size in the STAR file in Assets/TemplateMatching gives the desired outcome.
I had overlooked the STAR file in the TemplateMatching directory as the being the source of the metadata being used in creation of the Refinement Package.
Cheers!
